Biological target:
Hydrocortisone sodium succinate (Hydrocortisone 21-hemisuccinate sodium salt) has significant inhibitory effects on the IL-6 bioactivity with IC50 of 6.7 μM.
In vitro activity:
Steroidal anti-inflammatory drugs (SAIDs) exhibited significant inhibitory effects at 100 microM on the IL-6 bioactivity, and their inhibitory potencies were in the order of budesonide (IC50=2.2 microM)>hydrocortisone 21-hemisuccinate (6.7 microM), prednisolone (7.5 microM), betamethasone (10.9 microM)>dexamethasone (18.9 microM) and triamcinolone acetonide (24.1 microM). In vivo activity:
Two-way ANOVA revealed significantly increased plasma testosterone levels at 120 min (P ≤ 0.0037), 180 min (P ≤ 0.0016), and 240 min (P ≤ 0.0001) intervals in hydrocortisone treated monkeys. These results are suggesting an acute stimulation of plasma testosterone by RF9 compared to vehicle. Mean plasma testosterone after RF9 and vehicle administration in the hydrocortisone treated monkeys are shown in Figure 2.
