Blebbistatin is a myosin II inhibitor. Blebbistatin stabilizes the super-relaxed state in skeletal muscle. Blebbistatin exert antitumorigenic properties in human hepatocellular carcinoma cells by interfering with intracellular HuR trafficking. Blebbistatin is phototoxic to human cancer cells under exposure to blue light. Blebbistatin suppresses Ca(2+)-induced and "sensitized"-contraction of skinned tracheal muscles from guinea pig. Blebbistatin, a myosin II inhibitor, suppresses contraction and disrupts contractile filaments organization of skinned taenia cecum from guinea pig.

Biological target:
(-)-Blebbistatin is a selective inhibitor of the ATPase activity of non-muscle myosin II.
In vitro activity:

In vivo activity:
Compared with the CAL model group, mice in the Ble (blebbistatin) treatment group exhibited significantly reduced relative average absorbance for NMMHCIIA staining (Fig. 2A, B). Immunofluorescence analyses showed that NMMHCIIA expression was significantly lower in the endothelia of carotid arteries (green) in the sham group than that in the model group (p < 0.05). Moreover, blebbistatin intervention reduced the fluorescence intensity of NMMHCIIA staining (green) significantly (Fig. 2C, D). With regard to quantitative PCR, blebbistatin inhibited mRNA expression of NMMHCIIA in the CAL model (Fig. 2E).
