JNJ0966 is a potent and highly selective allosteric inhibitor of matrix metalloproteinase-9 (MMP-9), distinct from traditional active-site zinc chelators. It binds to a regulatory site on the MMP-9 zymogen, preventing its activation without affecting the catalytic activity of the mature enzyme. In enzymatic assays, JNJ0966 exhibited an IC₅₀ of approximately 440 nM for MMP-9 activation inhibition, while showing minimal to no inhibitory effect on other MMP family members such as MMP-2, MMP-1, or MMP-13 at concentrations up to 50 µM. In cell-based models, JNJ0966 reduced MMP-9–dependent extracellular matrix degradation and attenuated neuroinflammation in animal models, highlighting its potential therapeutic utility in MMP-9–driven pathologies with a favorable selectivity profile.

JNJ0966 is a highly selective MMP-9 zymogen inhibitor with an IC50 of 440 nM.
In vitro activity:
The efficacy of the inhibitor behaves in a dose-dependent manner, and this study determined that a 2-h pre-treatment with 20 µM of JNJ0966 could prevent the elongation of rat LECs that have been exposed to 6 ng/mL of TGF-β for 48 h.
