Biological target:
Spectinomycin HCl hydrate selectively targets the bacterial ribosome and interrupting protein synthesis. Spectinomycin HCl hydrate is also a noncompetitive inhibitor of td intron RNA with an Ki value of 7.2 mM-.
In vitro activity:
Neisseria gonorrhoeae (NG) increasingly resists antibiotics, prompting the search for new drugs. Spectinomycin and sanguinarine were tested against 117 clinical NG isolates. All isolates were susceptible to spectinomycin. Sanguinarine showed promising activity with minimum inhibitory concentrations (MICs) ranging from 2-64 μg/ml and a killing effect similar to spectinomycin over a 6-hour period. In vivo activity:
Spectinomycin displays a dose-dependent neuromuscular blocking activity on the rat sciatic nerve-gastrocnemius muscle preparation.
