Buspirone is an anxiolytic psychotropic drug of the azapirone chemical class. It is primarily used to treat generalized anxiety disorder (GAD). Unlike most drugs predominantly used to treat anxiety, buspirone's pharmacology is not related to benzodiazepines or barbiturates, and so does not carry the risk of physical dependence and withdrawal symptoms for which those drug classes are known.

Biological target:
Buspirone hydrochloride is an anxiolytic psychotropic drug, is used to treat generalized anxiety disorder (GAD).
In vitro activity:
Buspirone counteracted LPS-induced NO release, NOS2, IL-1β and TNF-α gene expression in WT cells, whereas it exerted limited effects in Drd3-/- or Htr1a-/- microglia. In summary, findings indicate that buspirone attenuates microglial polarization after LPS challenge. Reference: Cells. 2021 May 25;10(6):1312. https://pubmed.ncbi.nlm.nih.gov/34070429/
In vivo activity:
Exposure to IS increased the anxiety-/depression-like behaviors: it decreased the time spent in the open arm (OT) in the elevated plus maze (EPM) task and time spent in the light box (TL) in the light/dark transition (LDT) task and increased immobility times in the tail suspension test (TST) and forced swimming test (FST) (Fig. 1A–D). However, oral administration of buspirone significantly reduced IS-induced anxiety-/depression-like behaviors and suppressed IL-1β and TNF-α expression and NF-κB+/Iba1+ cell population in the hippocampus (Fig. 1A–G, Supplementary Fig. S1A).
