Genevant’s CL1 (also referred to as Lipid 10 or Lipid CL1) is an ionizable cationic trialkyl lipid commonly used as a key component in lipid nanoparticle (LNP) formulations for delivering nucleic acids such as mRNA or siRNA. CL1 is designed to remain largely neutral at physiological pH to reduce toxicity but becomes protonated under acidic endosomal conditions, promoting endosomal escape and efficient cargo release into the cytoplasm. While specific peer-reviewed quantitative data on CL1’s stand-alone bioactivity is limited due to its proprietary nature, preclinical studies and patents indicate that CL1-containing LNPs can achieve potent in vivo transfection efficiencies comparable to or better than earlier generation lipids like MC3, with mRNA delivery resulting in robust protein expression in the liver and other tissues at doses in the range of 0.05–0.5 mg/kg in rodent models.
