QXG-6442 is a potent and selective CK1α molecular glue Degrader. QXG-6442 demonstrates high proteome-wide selectivity and robust CK1α degradation capacity. QXG-6442 exhibited DC50= 5.7 nM and a Dmax of 90% in MOLT-4 cells after 5 hours of treatment at 1 µM. QXG-6442 selectively targets CK1α without affecting other CRBN neosubstrates. CK1α degradation by QXG-6442 was rescued in CRBN-knockout cells, confirming its CRBN dependency.
