GC-072 is the etiological agent of melioidosis and a Gram-negative bacterium. GC-072 is a potent, 4-oxoquinolizine antibiotic with selective inhibitory activity against bacterial topoisomerases. GC-072 exhibits in vitro potency against susceptible and drug-resistant strains of B. pseudomallei, as well as other dangerous pathogens such as Burkholderia mallei, Bacillus anthracis, Yersinia pestis, and Francisella tularensis. When administered orally in mice models mimicking melioidosis infection, GC-072 showed dose-dependent survival benefits, suggesting its potential as a first-line oral treatment option for melioidosis.
