Golcadomide (CC-99282) is a cereblon E3 ligase modulator (CELMoD) developed by Bristol Myers Squibb. It exhibits potent antitumor activity primarily through degradation of Ikaros (IKZF1) and Aiolos (IKZF3) transcription factors in B-cell malignancies. In preclinical studies, Golcadomide demonstrated sub-nanomolar DC₅₀ values for IKZF1 and IKZF3 in multiple myeloma and diffuse large B-cell lymphoma (DLBCL) cell lines, with DC₅₀ values typically in the range of 0.2–0.5 nM. It also induces apoptosis and inhibits proliferation in tumor cells by enhancing immune-stimulatory pathways.
