Biological target:
MIRA-1 can induce apoptosis in mutant p53 cells via restoration of p53-dependent transcriptional transactivation.
In vitro activity:
This study tested whether MIRA-1 could induce mutant p53-dependent growth inhibition in human SKOV ovarian carcinoma or H1299 lung adenocarcinoma cells carrying His175 or His273 mutant p53 under control of the tetracycline promoter. In all three cell lines expressing mutant p53 under control of the tetracycline-dependent promoter this study observed mutant p53-dependent growth suppression similar to that seen in Saos-2-His273 cells. In vivo activity:
This study sought to examine whether in vitro MIRA-1-induced MM cytotoxicity can be translated into effective in vivo anti-MM activity in SCID mice xenografted 8226 cells. Treatment with MIRA-1 significantly retarded tumour growth, with maximum tumour growth inhibition noted at day 15 (Figure 6A) (P<0.05). MIRA-1 treatment was also associated with improved survival, evidenced by first death at day 20 in control versus day 45 in treated group (Figure 6B) (P=0.007).
