Biological target:
Rubimaillin induces S-phase arrest of HepG2 cells, and increased intracellular reactive oxygen species levels. Rubimaillin induces DNA damage in HepG2 cells, as well as an increase in the expression of p-H2AX. Rubimaillin shows anti-cancer effect by inhibiting TNF-α-induced NF-κB activation. Rubimaillin enhances the osteogenic action of BMP-2 (bone morphogenetic protein 2) via the p38-Smad signaling pathway.
In vitro activity:
Rubimaillin was shown to have a strong inhibitory effect on HepG2 cells, inducing the S-phase arrest of HepG2 cells and increasing intracellular ROS levels. Comet assay demonstrated that Rubimaillin induced DNA damage in HepG2 cells. Additionally, Rubimaillin induced changes in cyclin A2 and CDK2. In vivo activity:
Rubimaillin ameliorated allergic airway inflammation by inhibiting Th2 response and macrophage polarization. Rubimaillin mitigated shrimp tropomyosin (ST)-induced elevation of arginase-1 expression and macrophage levels in the lungs and bronchoalveolar lavage fluid, respectively. In ST-stimulated mouse primary splenocytes, Rubimaillin notably inhibited IL-4 and IL-5 production and downregulated PARP1 and PAR protein expression.
