Biological target:
The biological target of suprafenacine is microtubules. It binds to the colchicine-binding site on microtubules, leading to destabilization and inhibition of microtubule polymerization.
In vitro activity:
In this in vitro study, Suprafenacine (SRF), a novel tubulin-destabilizing molecule, was identified through in silico screening. SRF binds to the colchicine-binding site on microtubules, inhibiting polymerization and leading to G2/M cell cycle arrest and subsequent cell death via a mitochondria-mediated apoptotic pathway. Notably, SRF demonstrated selectivity in inhibiting cancer cell proliferation and efficacy against drug-resistant cancer cells.
