ARP-101 is a selective MMP-2 inhibitor that displays ~ 600-fold selectivity over MMP-1 (IC50 values are 0.81 and 486 nM respectively). ARP101 induces autophagy-associated cell death in cancer cells. ARP101 treatment was highly effective in inducing the formation of autophagosome and conversion of LC3I into LC3II. Moreover, ARP101-induced autophagy was completely blocked in mouse embryo fibroblasts that lacked autophagy related gene 5 (ATG5(-/-) MEF). ARP101 inhibited α-MSH-stimulated melanin synthesis and suppressed the expression of tyrosinase and TRP1 in immortalized mouse melanocytes. ARP101 also induced autophagy in melanocytes.
