Epigallocatechin gallate (EGCG), also known as epigallocatechin-3-gallate, is the ester of epigallocatechin and gallic acid, and is a type of catechin. EGCG is the most abundant catechin in tea and is a potent antioxidant that may have therapeutic applications in the treatment of many disorders (e.g. cancer). It is found mainly in white tea, green tea and, in smaller quantities, black tea; during black tea production, the catechins are mostly converted to theaflavins and thearubigins, and theabrownins. It is also found in various vegetables, nuts, as well as carob powder at 109 mg per 100g.

Biological target:
(-)-Epigallocatechin Gallate is reported to inhibit EGFR signaling and thereby exert anticancer effects. (-)-Epigallocatechin Gallate (EGCG) is a glutamate dehydrogenase 1/2 (GDH1/2, GLUD1/2) inhibitor. Epigallocatechin-3-gallate induces oxidative phosphorylation (OXPHOS) by activating cytochrome c oxidase.
In vitro activity:
Caffeine and EGCG significantly increased F-actin level by 46.5% and 92.7%, respectively (Fig. 2B). In addition to immunofluorescence staining, Western blotting demonstrated the increased level of β-actin in caffeine-treated and EGCG-treated ECs when compared with the control cells (Fig. 2C). In consistent, quantitative data revealed that β-actin level was significantly enhanced by 53.4% and 76.0% by caffeine and EGCG, respectively (Fig. 2D). In vivo activity:
Western blot analysis showed increased iNOS and COX-2 expressions in tissues from vehicle treated rats 24 h, and three and five days after surgery, as compared to sham animals. EGCG administration reduced both expressions at all timepoints (Figure 7A,B). Additionally, PGE2 levels were significantly increased 24 h, and three and five days after surgery and EGCG administration reduced its levels (Figure 7C).
