Halobetasol propionate is a highly potent synthetic corticosteroid used topically for inflammatory skin conditions such as psoriasis and eczema. In vitro studies show that halobetasol binds GR with high affinity (IC₅₀ ≈ 0.4–0.8 nM), surpassing the potency of other corticosteroids like clobetasol. It suppresses pro-inflammatory cytokine expression, including IL-1β, IL-6, and TNF-α, in keratinocytes and fibroblasts. In vivo, halobetasol demonstrates rapid onset of action and marked reduction in erythema and plaque thickness in psoriatic models.
