Mandimycin is a macrolide antibiotic produced by Streptomyces species, belonging to the group of 16-membered macrolides. It exhibits potent antibacterial activity primarily against Gram-positive bacteria. In vitro studies show that Mandimycin A and B inhibit the growth of Bacillus subtilis and Staphylococcus aureus, with minimum inhibitory concentrations (MICs) in the range of 0.05–0.1 µg/mL. The antibiotic acts by inhibiting bacterial protein synthesis, likely through binding to the 50S ribosomal subunit, similar to other macrolides. It has also demonstrated modest cytotoxic activity in some mammalian cell lines, although its clinical development has not advanced. Structural analogs of Mandimycin are of interest in synthetic biology and medicinal chemistry due to their unique polyene macrolide scaffold and potential for modification.
