Trigriluzole, a tripeptide derivative of riluzole, exhibits neuroprotective and anticonvulsant activity by modulating glutamate neurotransmission. Preclinical studies show it reduces extracellular glutamate levels by enhancing glutamate uptake via EAAT2 transporters, with an EC50 of ~10 μM in astrocyte cultures. In rodent models of epilepsy, trigriluzole (10–30 mg/kg, oral) significantly decreases seizure frequency and neuronal hyperexcitability. It also demonstrates neuroprotective effects in neurodegenerative models, reducing oxidative stress and apoptosis.
