DDO-02005 is a potent and selective inhibitor of the Kv1.5 potassium channel, exhibiting an IC₅₀ of 0.72 μM. In vivo studies have demonstrated its efficacy in suppressing atrial fibrillation (AF) in a CaCl₂-acetylcholine-induced AF rat model and mitigating arrhythmias triggered by aconitine. Pharmacokinetic profiling in Sprague-Dawley rats revealed that, following intravenous administration at 1 mg/kg, DDO-02005 has a half-life of 3.23 ± 1.07 hours, a Cₘₐₓ of 90.23 ± 28.83 μg/L, and an AUC₀–ₜ of 178.42 ± 39.33 μg·h/L. Oral administration at 1.25 mg/kg resulted in a prolonged half-life of 6.25 ± 2.40 hours but significantly lower bioavailability, with a Cₘₐₓ of 1.27 ± 0.40 μg/L and an AUC₀–ₜ of 4.41 ± 0.69 μg·h/L, indicating poor oral absorption.
