CB-5339 is a VCP/p97 inhibitor with the potential for use in anti-cancer therapy. By binding to p97, CB-5339 suppresses ubiquitin-dependent protein degradation, leading to the accumulation of poly-ubiquitinated proteins in cells, which activates the ER-dependent stress response pathway, causing apoptosis in tumor cells overexpressing p97. As of 2022, CB-5339 has been undergoing Phase 1 clinical trials for use in treating R/R AML (acute myeloid leukemia) and MDS.
