Biological target:
Phosphatidylserine (PS) is an anionic phospholipid that is normally restricted to the inner leaflet of the plasma membrane bilayer of cells, appearing on the outer leaflet early in apoptosis and thus available for recognition by phagocytes. DPPS is a form of PS that contains the abundant long-chain (16:0) palmitic acid inserted at the sn-1 and sn-2 positions. It is commonly used in the generation of liposomes and other types of artificial membranes.
In vitro activity:
There is a critical size after which DPPS single shell vesicles are no longer stable. This study was able to infer a lower bound for the area dilation modulus for DPPS (252 pN/nm) and demonstrate a substantial free energy increase associated with vesicle rupture. In vivo activity:
A rat model of laser-induced choroidal neovascularization showed that annexin-DPPS-phytantriol (ANX-PS-Phy) cubosomes specifically targeted apoptotic cells in vivo. This study proposes that ANX-PS-Phy cubosomes are a potential candidate for ocular drug delivery for eye diseases.
