SL327 is a MEK inhibitor. SL327 blocks acquisition but not expression of lithium-induced conditioned place aversion: a behavioral and immunohistochemical study. SL327 had no affect on acquisition, expression, or extinction of EtOH-induced CPP in mice despite causing significant reduction of pERK levels in multiple brain regions. Additionally, although SL327 caused a generalized depression of locomotor activity, it did not prevent the development of EtOH-sensitization.

Biological target:
SL327 inhibits ERK1, MKK/p38, MKK4, JNK, and PKC at high concentrations (IC50s = >50, 21, >100, >100, >10 μM, respectively).
In vitro activity:
After experimental CNS hemorrhage, the breakdown of hemoglobin can cause iron-dependent neuronal damage, which heme oxygenase (HO) inhibitors may alleviate. U0126, SL327, and FR180204 reduced baseline HO activity by 35-50%, protecting against widespread neuronal injury caused by hemoglobin exposure. The results suggest that the protective effect of MEK and ERK inhibitors may be linked to a reduction in HO activity against heme-mediated neuronal damage. In vivo activity:
Mice were given SL327 before ethanol doses for acquisition or before post-conditioning tests for expression. SL327 significantly blocked the acquisition and expression of ethanol-induced CPP, but not CPA. Additionally, SL327 prevented the increase of pERK-positive neurons in specific brain regions. These findings suggest MEK's differential involvement in distinct behavioral outcomes during drug-induced place conditioning, influenced by the motivational sign and learning phase.
