In vivo, DMDHP-formulated LNPs showed robust luciferase expression in mouse liver and spleen tissues following intravenous injection, with luminescence signals comparable to or surpassing those of standard ionizable lipids. Additionally, DMDHP exhibits low cytotoxicity at working concentrations (<10 μg/mL), making it suitable for therapeutic development involving mRNA, siRNA, or CRISPR-Cas delivery.
