ISRIB is a potent PERK inhibitor.
In vitro activity:
The administration of ISRIB significantly inhibited the apoptosis of NSCs, and the flow results were consistent with the in vivo results (Figures 5(d) and 5(e)). The above results suggest that ISRIB administration alleviates the apoptosis of NSCs induced by hypoxic exposure. In vivo activity:
Alizarin red staining, calcium content, and ALP activity in the rat aorta were used to determine whether ISRIB could ameliorate VC progression. The increased calcium content and ALP activity in the calcified aorta were rescued by ISRIB treatment (Fig. 1B, C).
