pNZ-TMP is a TMP prodrug, and is the first small-molecule approach capable of detecting and controlling engineered cell-cell outputs. pNZ-TMP is also sterically “caged” TMP with 27-fold reduced binding affinity for reporter-DD protein fusions in dose response assays. The “caged” prodrug is enzymatically liberated by an activator-cell, restoring high affinity for the protein target, and can diffuse into the surrounding medium as shown by a media transfer experiment. If the liberated drug reaches a nearby receiver-cell in sufficient concentration, it can stabilize DD fusion proteins eliciting a protein signal output, thereby reporting an interaction.
