Biological target:
INCA-6 (Triptycene-1,4-quinone) is a cell-permeable NFAT inhibitor. INCA-6 specifically blocks targeting of NFAT(P) substrate to the calcineurin (CN) phosphatase site and is an effective inhibitor of CN-NFAT signaling.
In vitro activity:
INCA-6, which inhibits NFAT translocation into nuclei, suppressed the increase in osteocyte apoptosis in the compressive force-loaded parietal bones. Reference: JBMR Plus. 2020 May 26;4(7):e10369. https://pubmed.ncbi.nlm.nih.gov/32666020/
In vivo activity:
Lastly, as a proof of concept that NFAT-inhibition may be a viable chemotherapeutic strategy to address TNFα-induced retinal leukostasis, the effect of non-isoform-specific CN/NFAT inhibition was evaluated in vivo using the broad-spectrum NFAT inhibitor INCA-6. Intravitreal injection of TNFα (50 ng/ml) significantly induced retinal leukocyte adhesion compared to control after 6 hrs (p = 0.0023, Fig. 4), and co-injection of INCA-6 (25 μM) reduced this effect by 79.2% (p = 0.0089).
