NCI-65828, also known as N 65828, is an angiogenin (ANG) inhibitor. It functions by inhibiting ANG's ribonucleolytic activity, which is essential for its biological functions. Studies have demonstrated that NCI-65828 effectively reduces ANG activity, leading to decreased cellular proliferation in various cancer cell lines, including UROtsa, T24, and HeLa, with IC₅₀ values of 3.2 ± 0.8 μM, 1.3 ± 0.5 μM, and 1.9 ± 0.4 μM, respectively. Additionally, NCI-65828 has been shown to inhibit endothelial tube formation, a key process in angiogenesis, in human umbilical vein endothelial cells (HUVECs). In vivo studies have indicated that NCI-65828 can prevent prostatic intraepithelial neoplasia (PIN) formation in mouse models by inhibiting ANG's enzymatic activity without affecting its nuclear translocation.
