In vitro activity:
P-gp protein expression was substantially reduced by 36 ± 10% in hCMEC/D3 cells treated with 250 μM FAC for 72 h (Fig. 2a/b). To establish whether this was associated with a concomitant reduction in P-gp gene transcription, RNA was isolated from cells at various timepoints after FAC (ferric ammonium citrate)-treatment was initiated (as indicated in Fig. 2c) and MDR1 mRNA levels subsequently determined using RT-qPCR.
