Biological target:
Rhodblock 6 is an inhibitor of the Rho Kinase pathway. Rhodblock 6 directly inhibited Rok and its human ortholog ROCK I in a kinase assay.
In vitro activity:
This study discovered the molecules Rhodblock and identified their target as Rho kinase. Several Rhodblocks inhibited the correct localization of phosphorylated myosin light chain during cytokinesis. Rhodblocks differentially perturb Rho pathway proteins in cells and can be used to dissect the mechanism of the Rho pathway during cytokinesis.
