Description:

WARNING: This product is for research use only, not for human or veterinary use.

Rottlerin is an angiogenesis inhibitor and an inhibitor of protein kinase Cdelta (PKCdelta) and calmodulin kinase III. Rottlerin as a novel chemotherapy agent for adrenocortical carcinoma. Rottlerin induces cyclooxygenase-2 upregulation through an ATF4 and reactive oxygen species-independent pathway in HEI-OC1 cells. Rottlerin exerts its anti-tumor activity through inhibition of Skp2 in breast cancer cells. Rottlerin suppresses growth of human pancreatic tumors in nude mice, and pancreatic cancer cells isolated from Kras(G12D) mice.

Product Data
Biological target:
Rottlerin has IC50 values for PKCδ of 3-6 μM, PKCα,β,γ of 30-42 μM, PKCε,η,ζ of 80-100 μM. Rottlerin acts as a direct mitochondrial uncoupler, and stimulates autophagy by targeting a signaling cascade upstream of mTORC1. Rottlerin induces apoptosis via caspase 3 activation.
In vitro activity:
The effects of rottlerin on CYP2C8 inhibition and EET formation insinuate further exploration for cancer therapy. Rottlerin showed highly potent and selective CYP2C8 inhibition (IC50 < 0.1 μM) in human liver microsomes. Reference: Chem Biol Interact. 2023 Aug 1;380:110524. https://pubmed.ncbi.nlm.nih.gov/37146929/
In vivo activity:
In a highly aggressive mouse mammary carcinoma model, rottlerin has significant potential to augment the anti-metastatic efficacy of paclitaxel via impeding EMT activation along with attenuating its treatment-associated toxicological alterations.
