CHF5022 is a flurbiprofen analogue that inhibits amyloid-beta 42 (Aβ42) secretion, exhibiting an IC50 of 92 μM in vitro. Notably, CHF5022 demonstrates a threefold greater potency in reducing Aβ42 secretion compared to R-flurbiprofen, which has an IC50 of 268 μM. Importantly, CHF5022 does not inhibit cyclooxygenase (COX) enzymes at concentrations up to 300 μM, distinguishing it from R-flurbiprofen, which retains significant COX-1 and COX-2 inhibitory activity within this concentration range. Additionally, CHF5022 exhibits modest activity against Aβ40, achieving a reduction of 22.2% ± 4.8% at a 100 μM concentration without inducing cytotoxicity.
