Moxidectin is a semisynthetic derivative of nemadectin which is produced by fermentation by Streptomyces cyano-griseus. This Streptomyces species was discovered in a soil sample from Australia in the late 1980s collected by an agronomist working for the American Cyanamid company.

Moxidectin(ProHeart 6; CL301423; Cydectin) is an anthelmintic drug which kills parasitic worms (helminths), and is used for the prevention and control of heartworm and intestinal worms.
In vitro activity:
The main objective of this study was to explore autophagy induced by MOX in glioma U251 and C6 cells, and the deep underlying molecular mechanisms. The effect of MOX on the induction cell autophagy in glioma cells was examined by TEM and GFP-LC3 transfection. As shown in Fig.1A, TEM revealed obviously characteristic autophagosomes in U251 and C6 cells treated with MOX but not in control cells. Arrows indicated autophagosomes containing intact and degraded cellular debris. The induction of autophagy was determined by GFP-LC3 transfection. As shown in Fig.1B, abundant LC3 puncta was observed in 48 h MOX-treated cells compared with untreated U251 and C6 cells. In vivo activity:
Since GM (gut microbiota) plays a role in all these conditions and MOX may affect at least some of these behaviors (e.g. alcohol intake), the effects of MOX on GM in WKY rats were examined. Adult male Wistar Kyoto (WKY) rats (5/group) were injected intraperitoneally (i.p.) once daily for 7 days with MOX (2.5mg/kg), or saline as control group.MOX significantly enriched Lactobacillaceae, Microbacteriaceae, Rhodospirillaceae and Pelagicoccaceae by 3, 4, 4, and 4-fold, respectively. Deferrribacteraceae, Erysipelotrichaceae and Thermovenabulum were reduced by 8, 2.5, and 3.5- fold respectively, compared to the control group at the family level [Figure 6]. MOX depleted 11 species, including 2 species of Ruminoccus, which are positively associated with severity of irritable bowel syndrome, and 4 species of Provettela, which are closely associated with depressive-like behavior. In summary, these findings indicate that chronic administration of MOX results in significant increases in the levels of probiotic bacteria (e.g. Lactobacillus, Bifidobacterium and Butyrivibrio), and significant decreases in opportunistic pathogens (e.g. Provettela and Mucispirallum) in WKY rats, a putative animal model of depression.
