CASIN is an inhibitor of Cdc42 GTPase, a small GTPase of the Rho-subfamily that plays important roles in cytoskeleton organization, cell cycle progression, signal transduction, and vesicle trafficking. CASIN generated a rejuvenated phenotype of Hematopoietic stem cells (HSCs), reversing the aging-related and polarity phenotype of aged HSCs to that of young HSCs in vivo.

Biological target:
CASIN is a selective GTPase Cdc42 inhibitor with IC50 of 2 uM.
In vitro activity:
In addition, CASIN was studied to see how it affects cell migration, apoptosis and cell cycle of endothelial cells. CASIN upregulates key endothelial marker genes and downregulates a number of cyclin dependent kinase inhibitors. These findings suggest that CASIN could be further tested in the development of artificial vascular systems and vascular gene editing technologies. Furthermore, these findings may have potential to contribute to the development of alternative treatment methods for diseases that cause endothelial damage. In vivo activity:
It was demonstrated that systemic treatment of aged (75-week-old) female C57BL/6 mice with a Cdc42 activity-specific inhibitor (CASIN) for 4 consecutive days significantly extends average and maximum lifespan. Moreover, aged CASIN-treated animals displayed a youthful level of the aging-associated cytokines IL-1β, IL-1α, and INFγ in serum and a significantly younger epigenetic clock as based on DNA methylation levels in blood cells. Overall, our data show that systemic administration of CASIN to reduce Cdc42 activity in aged mice extends murine lifespan.
