Methylprednisolone acetate, a prednisolone derivative, is a corticosteroid hormone.
In vitro activity:
In addition to its potent anti-inflammatory and antioxidant properties, this study recently discovered a selective antiapoptotic effect of MP (methylprednisone) on oligodendrocytes via the activation of the glucocorticoid receptor (GR) and the upregulation of bcl-X(L), a splicing isoform of the bcl-x gene. The GR and STAT5 complex is present on the STAT5-binding site of the bcl-x promoter region in oligodendrocytes; the overexpression of an activated form of STAT5 prevents alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid-induced oligodendrocyte cell death; and this prevention is lost when the STAT5 gene is knocked down. In vivo activity:
This study aimed at evaluating the effect of methylprednisolone (MPA) on messenger ribonucleic acid (mRNA) expression levels in immature ovine knee joint tissue explants following interleukin (IL)1β induction and to assess responsiveness of the explants. Methylprednisolone acetate treatment significantly suppressed MMPs consistently across the cartilage (MMP1, MMP3, and MMP13), synovium (MMP1 and MMP3), and IPFP (MMP13) (all p < 0.05).
