JAB-3312 is a potent, selective, and orally bioavailable small-molecule inhibitor of the protein tyrosine phosphatase SHP2 (Src homology region 2-containing protein tyrosine phosphatase 2), a key regulator of the RAS/MAPK signaling pathway. By inhibiting SHP2, JAB-3312 disrupts oncogenic signaling driven by receptor tyrosine kinases and RAS mutations. In biochemical assays, JAB-3312 demonstrates nanomolar potency against SHP2, and in cell-based assays, it inhibits downstream ERK phosphorylation, confirming effective pathway suppression. Preclinical studies have shown that JAB-3312 inhibits tumor growth in xenograft models of SHP2-dependent cancers, including EGFR-mutant and KRAS-mutant tumors, particularly when used in combination with other agents such as MEK or KRAS G12C inhibitors.
