BBDDL2204 is an EZH2 inhibitor. BBDDL2204 exhibited excellent EZH2Y641F enzymatic inhibitory activity with an IC50value of 2.5 nM. In addition, BBDDL2204 significantly reducedH3K27Me3 levels and displayed much enhanced potency in inhibiting the cell growth of KARPAS422 and Pfeiffer cells than1. Furthermore, kinetic experiments demonstrated BBDDL2204 binding to EZH2 in a SAM-noncompetitive mode.
