​MC3343 is a potent, non-nucleoside DNA methyltransferase (DNMT) inhibitor, exhibiting IC₅₀ values of 5.7 μM for DNMT1 and 1.7 μM for DNMT3a in cell-free assays. Compared to earlier inhibitors like RG-108 and SGI-1027, MC3343 demonstrates enhanced potency against DNMT1. In osteosarcoma (OS) models, particularly in Saos-2 cells, it effectively suppresses proliferation and promotes osteoblastic differentiation. Treatment with MC3343 leads to dose- and time-dependent reductions in DNMT1, DNMT3a, and DNMT3b protein levels. In vivo studies reveal that MC3343 synergizes with chemotherapeutic agents such as doxorubicin and cisplatin, resulting in diminished growth of patient-derived xenograft OS tumors and induction of differentiation
