Desmethylglycitein (DMG), a minor isoflavone predominantly found in fermented soy products, exhibits notable estrogenic and antioxidant activities. In vitro, DMG shows significant affinity for estrogen receptor β (ERβ), with a relative binding affinity of approximately 14% compared to estradiol, favoring ERβ over ERα, which suggests potential for tissue-selective estrogenic effects. DMG also demonstrates antioxidant properties, evidenced by its ability to inhibit lipid peroxidation in rat liver microsomes and scavenge DPPH radicals with an IC₅₀ of ~13 μM. Additionally, DMG has been shown to inhibit proliferation of MCF-7 breast cancer cells with an IC₅₀ around 30–40 μM, indicating weak antiproliferative activity that may be mediated through modulation of estrogen signaling pathways.
