AZD4144 (also known as AZD04144) is a potent, selective, brain-penetrant inhibitor of the NLRP3 inflammasome. Preclinical studies have shown that AZD4144 effectively blocks NLRP3-mediated IL-1β release in vitro with an IC₅₀ in the low nanomolar range (reported around 20–30 nM in human PBMCs). In rodent models, AZD4144 demonstrated robust target engagement and significant reduction of IL-1β and IL-18 levels in the brain and periphery, indicating its potential for treating neuroinflammatory conditions such as Alzheimer’s disease and other CNS disorders involving aberrant NLRP3 activation.
