AVS-100 is a novel histone deacetylase (HDAC) inhibitor designed to selectively inhibit class I and II HDAC isoforms, thereby inducing hyperacetylation of histone proteins and reactivating silenced tumor suppressor genes. In preclinical studies, AVS-100 demonstrated potent inhibition of HDAC enzyme activity with IC₅₀ values in the low nanomolar range (e.g., ~5–15 nM for HDAC1 and HDAC2). In cellular assays, AVS-100 significantly increased acetylated histone H3 levels and induced apoptosis in various cancer cell lines, including colon and breast cancer models, with GI₅₀ values ranging from 50 to 200 nM. In vivo, AVS-100 showed dose-dependent tumor growth inhibition in xenograft models, achieving up to 70% tumor growth suppression at tolerated doses, with minimal systemic toxicity observed.
