Y-30141 is a compound closely related to the widely used Rho-associated coiled-coil forming protein serine/threonine kinase (ROCK) inhibitor Y-27632. Y-30141 was examined for its inhibition mechanism and profile of actions. Y-30141, like Y-27632, inhibited the kinase activity of both ROCK-I and ROCK-II in vitro through competitive binding to the catalytic site, with affinities for ROCK kinases 20 to 30 times higher than for other Rho effector kinases.
