AOH1160 is a potent caPCNA inhibitorr. AOH1160 exhibits selective cytotoxicity against various cancer cell lines—including neuroblastoma, breast cancer, and small-cell lung cancer—with IC₅₀ values ranging from 0.11 to 0.53 μM, while demonstrating minimal toxicity (up to 5 μM) toward nonmalignant cells such as human PBMCs, mammary epithelial cells, and small airway epithelial cells. Mechanistically, AOH1160 disrupts DNA replication, impedes homologous recombination-mediated DNA repair, induces cell-cycle arrest, and promotes apoptosis via caspase-3 and caspase-9 activation. Notably, it enhances the sensitivity of cancer cells to cisplatin, with a combination index of approximately 0.55, indicating synergistic effects.
