Biological target:
GW6340 is an intestinal-specific LXR agonist. To evaluate the effect of LXR activation specifically in small intestine on mRCT, wild-type mice were treated with either intestinal-specific LXR agonist (GW6340) or systemic LXR agonist (GW3965). Initial studies using the systemic LXR agonist GW3965 showed no promotion of macrophage reverse cholesterol transport (mRCT) in mice lacking LXR expression in the liver or intestine. Evaluating the effect of LXR activation specifically in the small intestine, both intestinal-specific (GW6340) and systemic (GW3965) LXR agonists significantly increased sterol excretion in feces. However, when assessing the necessity of macrophage LXR activation, mice with LXR alpha/beta knockout macrophages showed significantly reduced mRCT promotion with GW3965 treatment, highlighting the importance of macrophage LXR activation in this process.
