BMS-615, also known as TLR7/8/9-IN-1, is a small-molecule inhibitor of Toll-like receptors (TLRs), particularly targeting TLR7 and TLR9, which are involved in innate immune responses and the pathogenesis of autoimmune diseases such as systemic lupus erythematosus (SLE). In preclinical studies, BMS-615 potently inhibited TLR7/9-mediated cytokine production (e.g., IFN-α, IL-6) in human peripheral blood mononuclear cells (PBMCs), with IC₅₀ values in the low nanomolar range (typically around 10–50 nM depending on the assay). The compound demonstrated good selectivity over other TLR family members and showed efficacy in murine models of lupus, reducing disease biomarkers and inflammation.
