Thalidomide-Photoswitch3-NH2 hydrochloride is a vital component in the synthesis of PHOTACs, which are PROTACs whose reversible activation is governed by different wavelengths of light, being rendered inactive in the dark and activated to the cis isomer by irradiation at 390 nm. At wavelengths above 450 nm, the PHOTACs are reversibly inactivated. The conjugate, which consists of a Cereblon (CRBN)-recruiting ligand, an azobenzene photoswitchable crosslinker, and a pendant amine that reacts with an acid on the target warhead, was used in the preparation of PHOTAC-I-10 and PHOTAC-II-6.
