HG381 is a potent and selective STING (stimulator of interferon genes) agonist, designed to activate the innate immune response via the cGAS-STING pathway. In preclinical studies, HG381 has demonstrated strong STING activation in human peripheral blood mononuclear cells (PBMCs), with an EC₅₀ in the low nanomolar range (~5–10 nM), leading to robust induction of type I interferons such as IFN-β and downstream cytokines like TNF-α and IL-6. In murine models, intratumoral administration of HG381 significantly enhanced antitumor immunity, resulting in tumor growth inhibition and increased infiltration of CD8⁺ T cells. Pharmacodynamic assays confirmed dose-dependent STING pathway activation, with upregulation of key markers such as p-TBK1 and IRF3.
