JC744 is a chemical probe and is responsible for stabilizing p53-Y220C in-vitro (Kd = 320 nM). P53 is a tumor suppressor that often becomes inactivated in human cancers, with Y220C being the most prevalent p53 mutant. Destabilization caused by the mutation, a result of a pocket at the surface of the DNA-binding domain, leads to rapid aggregation and denaturation.
