JTE-051 is a potent inhibitor of interleukin-2-inducible T cell kinase (ITK), a key enzyme involved in T cell receptor signaling. It inhibits ITK enzymatic activity with an IC₅₀ of approximately 2.2 nM. In cellular assays, JTE-051 suppresses T cell activation and cytokine production, notably reducing IL-2 and IFN-γ secretion in stimulated human peripheral blood T cells. In vivo, JTE-051 demonstrates strong anti-inflammatory effects in several animal models. For instance, in a rat collagen-induced arthritis (CIA) model, oral administration of 10 mg/kg/day significantly suppressed paw swelling and joint histopathology scores. These results support JTE-051’s therapeutic potential in T cell–mediated autoimmune disorders such as rheumatoid arthritis and psoriasis.
