JYQ-42 is a potent allosteric inhibitor of SIRT6. JYQ-42, the most efficient SIRT6-specific allosteric noncompetitive inhibitor, selectively targets SIRT6 and effectively inhibits the deacetylation activity of SIRT6 through several critical residue interactions. The IC50 value of JYQ-42 is 2.33 ± 0.17 μM.
