QLS-81 is a novel Nav1.7 channel blocker (IC50 at 3.5±1.5 μM). QLS-81 effectively inhibits Nav1.7 channel activity in a concentration-dependent manner. Electrophysiological assays revealed that QLS-81 significantly reduces the peak current of Nav1.7 channels, indicating its potential to diminish neuronal excitability associated with pain transmission. Additionally, QLS-81 was shown to enhance the slow inactivation of Nav1.7 channels, further contributing to its inhibitory effects. QLS-81 holds promise as a therapeutic agent for managing neuropathic pain, a condition often resistant to conventional analgesics. By selectively inhibiting Nav1.7, QLS-81 may offer pain relief with potentially fewer side effects compared to non-selective sodium channel blockers.
