Biological target:
SSO inhibits fatty acid transport into cells. SSO is a potent and irreversible inhibitor of mitochondrial respiratory chain.
In vitro activity:
This study used SSO-treated CHO cells expressing human CD36 to identify oleate modification of Lys-164, a key site in the FA-binding domain. SSO also inhibited oxLDL uptake by macrophages, suggesting competition for the CD36 binding pocket. These findings highlight the significance of Lys-164 in CD36 function and provide insight into its inhibition mechanism. In vivo activity:
In a model of diabetic hearts, SSO reduced fat oxidation but also lipotoxicity, by targeting the primary step in the fatty acid metabolism pathway. SSO infusion decreased the fatty acid oxidation rate by 29% and myocardial triglyceride concentration by 48% compared with untreated diabetic hearts. Infusing SSO into diabetic hearts increased rate pressure product by 60% during hypoxia and by 32% following reoxygenation, restoring function to control levels.
